  Dearest Parents,

  Life is proceeding reasonably well. It rained hard last night and today was extremingly beautiful. It was clear, cold, windy, clean, and sunny with blotches of white cloud. Though I could ill afford the time, I drove to Claremont to get our sandals. He did not have the leather so they are not ready. Next week I have finals. Boy.

  Walter has been coming and working, I guess. The Hughes moved out Monday. Sue has not shown.

  It is a shame that you did not apply a good bit of pressure on Bob to send Mariette out here for Christmas. It is really much better that she come here, but it will take persuasion. Work on it. I think it would be worthwhile to offer to Mariette to pay her way out here, if Bob says no. Meaning I could offer to Mariette to come out. She has her parents in a tight spot, where she can do as she pleases without fear of extensive repercussion. All they can do is cut off her support which cancels school and so they lose the big fight anyway.

  Oh Bob told her he was worried that all I wanted was to sleep with her & that he wanted her to finish college. I wrote and said I sure as hell did not have to marry a girl to get a mistress & being unmarried had the additional spice of variety. I was used to girls supporting me. (These comments are not true exactly; I could have a mistress but it would not be much fun, and variety ain't so hot afterall.) I also said if she preferred to finish college there was little I could do and [tant pis.?] I could not guarantee eventual marriage because life is not like that, and I would be unhappy. Anyway, her bad letter arrived a week ago Monday. The effect of her Pa has worn off and she is all love and kisses now.

  How are you? Today I received your letter from Washington. Very interesting. On Keith's floating school there would be plenty of time to study. It is boring at sea when there is nothing but water and air and the novelty of being on a ship would wear off quickly. Say hello to [?], Anita, Daddy, and all various and sundry children that may be there. Can Petey walk yet? My roommate and I are buying Pierre a record for Xmas. $2 each. He is also getting one for me, so when I am home next weekend I will get something for him. Do you think that I should apply for the University of Chicago next year? Keith thinks that it would do me more harm than good to stay here. Tony Converse thinks I am a little young, but most faculty members with whom I have conversed on the subject (excluding Toe. C. and M. C.) think that I should if I want. Of course, I would probably never get home, but then you come to Chicago often and we have many friends there. I think it might be worth doing. How is Boston? Do you like it there? My god, I must go to bed.

  Subject: Present status of the study of amino acid sequence in gelatin

  About 3 months ago we began to use the ion exchanger, Dowex-50, and the methods of Moore and Stein in an attempt to isolate peptides from partial hydrolysates of gelatin. In our initial experiments, we compared the results which were obtained when a partial hydrolysate and a complete hydrolysate of gelatin were chromatographed under identical conditions. Despite the arbitrary set of conditions which was used, the results were very gratifying: the chromatogram of the partial hydrolysate showed eleven zones which either were absent from the chromatogram of the complete hydrolysate and thus presumably were peptides or had characteristics indicating at least the presence of peptides.

  The partial hydrolysate which was used in these experiments had been obtained by refluxing gelatin in 6 N hydrochloric acid for half an hour. These conditions are very different from those of Grassmann and Riederle who isolated lysyl-prolyl-glycine from a hydrolysate prepared by heating at 37º in 3.6 N hydrochloric acid for 28 days. Recently, we have been duplicating their hydrolytic conditions and have been interested to find that the mixture of peptides so produced is very similar to that of our initial experiments.

  Up to the present time, the scale of our experiments has been so small that further work on the peptides which have been isolated can only e preliminary in scope. However, one peptide zone has been subjected to further study and has been found to contain threonylglycine in amount roughly equivalent to 10% of the theronine in gelatin.

  The results of this preliminary investigation are very encouraging and permit us to arrive at a tentative program which will make the best use of our present staff.

  (1) Of most immediate interest is an attempt to improve the separation of the observed peptide zones by variation of the conditions on the ion exchange columns. Each chromatogram requires one to two weeks for completion and Mr. Honnen and I can run what amounts to about one and a half chromatograms at a time. This study of the effect of variation in conditions will require 6 to 8 weeks to complete and even then will be far from thorough.

  (2) While the above work is in progress, Dr. Green and Miss Michelson will make a preliminary study of the small amounts of the various peptide zones which have been isolated in order that we may have an insight into the complexity of each and be able to decide which are most amenable to further productive study. It will probably not be possible to complete these preliminary studies during the 6 to 8 weeks which will be used to try to improve the separation of the peptide zones.

  (3) It is also desirable for us as soon as possible to begin experiments with paper chromatography. Paper chromatography may prove to be valuable in two ways: as a means of separating peptide mixtures which may be well present in the various peptide zones and also as a method for identifying the constituent amino acids in any peptide which may be isolate. The letter can be done by the DNP technique but confirmatory evidence from paper chromatography would be valuable. The extent to which we may be able to venture into paper chromatographic work will depend to a large extent upon the available space. At the present time, there is little space available in either of the laboratories and what is available may not be suitable for paper chromatographic work.

  (4) After we have completed the study which is projected in (1), larger chromatographic columns will be prepared and at least 10 to 20 times the amounts of peptides which are now available will be isolated. The preparation of ion exchanger for a large column will be a major task.

  (5)The larger quantities which will be isolated in (4) will then be subjected to exhaustive study. The complexity of the mixture in any pepetide zone will determine the amount of time which will be required to complete a study of it but it may be anticipated that for each zone the full time of one person may be needed for as little as two or three weeks or as much as six months.

  On the basis of the results which we have already obtained, I feel that we should be able to make a contribution to the structure of gelatin and of collagen but the rate of progress will certainly depend upon the size of our group and space and facilities which we can use. I have been wondering whether it might be possible to obtain financial support to maintain or increase the present group from industrial concerns which are interested in gelatin and collagen. I am thinking of such manufacturers as Wilson and Company, who produce the gelatin for OPG, Knox Gelatin, leather companies, and the like. If no support could be obtained from such sources, perhaps application should be made for a grant from the Research Corporation.

  In answer to your letter of 7 December, I am glad to give you my assessment of Julian M. Sturtevant.

  I have known Dr. Sturtevant for a dozen years, and I have read his papers, and inspected some of his apparatus. It is my opinion that he is an extraordinarily able investigator. He is meticulously careful in the design and construction of apparatus for his experimental work, and in the execution of the experiments. I feel that his experimental results can be relied on with as great confidence as the results of any other worker in this general field.

  Dr. Sturtevant is a careful scientist who makes sound contributions to the body of scientific knowledge. His work is of such a nature that it does not attract great attention; nevertheless I consider that it is valuable. Dr. Sturtevant has, in my opinion, a fine personality, and, although I have not observed him as a teacher, I would expect him to be a very good one.

  In summary, I would say that I believe that Dr. Sturtevant's abilities and achievements are such as to justify his promotion to Professor of Chemistry.

  Silicon compounds

  Would you write out a brief statement about work on electron diffraction in relation to the nature of the silicon- oxygen bond. Perhaps it would be enough for you to sketch out briefly the investigations on silicon compounds that have been made in our laboratories by the electron diffraction method, the incomplete studies (Si2Cl2 and any others), and the studies that you would propose to make. Anything that would throw light on the nature of silicon-oxygen bonds would be worth while. If you sketched this out I could prepare a letter to Mr. Belknap.

  What I suggest is that a post-doctorate man be obtained, who would put in only part of his time on silicon-oxygen compounds themselves, and part of his time on what may be called related substances. I hope that a grant could be obtained that would cover part of the cost of building the apparatus, too.

  Dear Henry:

  Thanks very much for your Christmas letter. Also, Ava Helen and I were happy to have the Christmas card that you and Edith sent us. We have not sent out any cards for several years, because of our being so busy, especially along before Christmas.

  I am sorry that you have been laid up with the flu. Should you not be getting another man in the office, to help you out - and especially to be available, together with Jim, when you take your sabbatical year?

  I shall look forward to receiving the batch of new-type cases, and also some of the 1952 digests. I thank you for the honorarium, which surprised me when it arrived last week. You know that I am glad to do anything that I can for you and the Foundation.

  We have three of the children at home with us for the holidays, along with two (at times three) other young people - friends of Linda down from Reed College. Ava Helen and I are feeling well, but I think that she is being kept too busy, by the house full of people.

  Tomorrow we are going down to the beach to collect marine animals, in order that I may prepare some specimens of muscle protein. An extraordinary situation has arisen. Twenty-five years ago some investigators in Germany dried a piece of muscle and made an x-ray photograph of it; this had been done scores of times before, and the x-ray photographs that had been obtained had always been the same - rather fuzzy, uninteresting diagrams - but this preparation gave a striking photograph, such as is obtained from a crystalline material. A similar crystalline photograph was obtained from another preparation in Switzerland ten years ago, and Bear, in Schmitt's laboratory, has made two samples of muscle that give the crystalline pattern. Corey and I discussed the photographed obtained by the Swiss workers, and described their sample as crystallized muscle. Bear published a letter in Nature last month, in which he states that he does not think that the crystalline material is muscle at all. It is hard to imagine what he thinks it is, because it is evident that the crystalline material makes up most of the sample, which was just a piece of muscle that had been dried. Corey and I feel that this crystalline pattern is extremely important, because of the large amount of information that it probably will provide (and in part already has provided) about the struc-

  ture of proteins, and even about the nature of the phenomenon of muscular contraction. We have the original sample of the Swiss investigators, Lotmar and Picken(Picken is an Englishman), but it no longer is crystalline - instead it gives the ordinary fuzzy muscle diffraction pattern.

  I estimate that some hundreds of samples of dried muscle have been made by investigators during the past thirty years and subjected to x-ray diffraction. Four of these samples have turned out to be crystalline. Nobody knows what the conditions are that produce the crystalline material. Accordingly I propose to make some hundreds of samples of dried muscle, in various ways, and to make x-ray photographs of all of then, with the expectation that in a month, or two months, or three months, or perhaps six months a sample of crystalline muscle will turn up. He can then make better x-ray photographs than have been obtained by the earlier investigators, and presumably derive some valuable information about protein structure and muscle structure from them. The four samples of crystallized muscle that were obtained by earlier investigators are from four different animals - frog, mussel, pectin, and squid. These are all cold blooded animals, and accordingly we are planning to get samples of muscle form cold blooded animals to begin with, and to try some mammalian muscle samples later on.

  The Royal Society is arranging a Conference on Protein Structure. I stated at first that I could not attend, because of the trip around the world. Now I have agreed to attend, and the date has been set for May 1, rather than April 24, in order to permit me to come to the Philosophical Society meeting and then to attend the Conference. Ava Helen and I are now planning to go to the Conference, and to take a vacation during May and June, and then to attend an International Conference on Biochemistry in July, which is to be held in Paris. We shall then come back home, about August 1.
