    Dear Mr. Boyer

    In answer to your letter of 20 February, I am glad to comment on the questions that you asked.

    1. I think that chemical intervention to correct enzyme abnormalities or deficiencies will become a practical clinical procedure in the not very distant future. For example, in the case of a hereditary enzyme abnormality or deficiency that causes trouble because of the piling up of the substrate that ordinarily is converted into other substances by the enzyme, the treatment might involve simply the decrease in amount of the substrate ingested by the patient. This treatment is already in use: for example, milk is removed from the diet for galactosemics, and phenylalanine is removed from the diet for phenylketonurics.

    I enclose a reprint of a paper in which I have suggested that within fifty years it will be possible to make synthetic substances with enzyme activity, and to use these artificial enzymes in the control of diseases resulting from abnormal or deficient enzymes.

    Another possibility is that an abnormal enzyme manufactured by a patient because of a genetic abnormality may be made effective through the combination with a simple chemical substance.

    2. I do not have any special knowledge about clinical work on chemical intervention to rectify metabolic abnormalities due to enzyme change from genic mutation.

    3. I think that it may well be possible in some cases to make clinical use of transduction. I think that the probability of successful use of transduction is especially high in the case of diseases in which, because of genic abnormality, the patient fails to manufacture an enzyme, or manufactures an abnormal enzyme that will not work. It might be that in some cases modification of cells by transduction, with use of deoxyribonucleic acid, or implantation of cells, may be done in such a way that a satisfactory amount of the enzyme is manufactured by the patient.
