  You may be interested to know that in New York Dr. Fajans met me at a reception and attacked me with great vigor, about the report that I rendered to you of an article that he and another man had submitted for publication in the Journal of the American Chemical Society, and that you had sent me, in order that I might serve as a referee. My report to you as a referee was a confidential one, but Dr. Fajan stated that by accident your office had included in a letter to him the signed letter that I had sent to you. You may remember that I had stated my opinion of this article frankly, without going to the trouble of phrasing the remarks in the polite way in which they would have been phrased (although the content might well have been the same) if I had been making a report directly to Fajans.

  Dear George:

  I am sorry that I haven't anything to say in answer to your letter about mechanism by which bio-electric potentials are maintained in the living cell. I haven't done any thinking about this field, nor have I read the literature about it.

  May I ask if you have made any tests of methods for preventing crises in sickle cell anemia? It seems to me that the use of carbon monoxide therapy could be tested out in the hospital. I think that 200 ml of carbon monoxide could be safely administered to an adult, and corresponding amounts, proportional to hemoglobin content of the individual, to children. Also, Dr. Itano has suggested that a methemoglobin produced by the administration of sodium nitrite would be effective in preventing crises. It would be necessary to determine by experiment the dosage of sodium nitrite required to keep about one fifth of the hemoglobin in the oxidized (methemoglobin or ferrihemoglobin) state. Even though the pharmacological factor for carbon monoxide and for nitrite is not a large one, it might well be that a treatment of this sort would have some value.

  Dr. Itano has just written a paper describing a third abnormal form of hemoglobin, associated with hereditary hemolytic anemia. This abnormal form has the same electrophoretic properties as sickle cell anemia hemoglobin, but it does not cause the cells to sickle, and its solubility properties are different.

  We have not been sending any sickle cell blood recently, as we were not certain that you wished to have the shipments continued. If you do, we shall be very happy to arrange this as we have in the past.

  I am very much interested in your polypeptides. I think that it would be well worth while for us to make an x-ray investigation of them.

  We would be very glad to study the compound composed of C-allyl-glycine units, and I suggest accordingly that you send us a sample of this substance. A 250 mg. sample, mentioned in your letter, would be satisfactory.

  I do not think that we should ask you to synthesize the D,L-leucine polypeptide, because D,L polypeptides are not well suited to x-ray investigation.

  Let me thank you for calling my attention to the availability of your materials. We are very anxious to the detailed structures of some synthetic polypeptides, and because of the element of chance involved in the structure determinations we want to investigate as many different substances as possible.

  Dear Dr. Ravdin:

  I have been interested in the problem of plasma substitutes or plasma extenders for a number of years, and during the last two years my concern about this problem has become greater and greater. I am writing now to ask if it might be possible for me to talk with you about it during my visit in Philadelphia early next month.

  In 1942 the Committee on Medical Research made a contract with the California Institute of Technology for the development of a plasma substitute. The initiative for this work come from Dr. Dan H. Campbell, Professor of Immunochemistry in our Division of Chemistry and Chemical Engineering. Work was done from 1942 to 1946, at first with me as official investigator, and then with Professor Campbell as official investigator. The product that we developed, Oxypolygelatin, is obtained by the chemical treatment of gelatin, in such a way as to improve its properties for transfusion. This product is covered by patent, with Professor Campbell, Dr. J. B. Koepfli (now Science adviser to the Secretary of State), and me as inventors.

  We had thought that our job was done in developing a product that seems to us to be very good, and we had expected that the commercial development of the material would be taken care of by other people - we are essentially basic scientists. However, about two years ago we realized that our Oxypolygelatin was being ignored by the authorities, and we succeeded in obtaining a Public Health Service grant for its further study. We have been continuing with this work.

  Professor Campbell and I have the feeling that Oxypolygelatin is not being considered sufficiently seriously, in comparison with other proposed plasma extenders. We feel great concern about this situation.

  First, let me say that it is my own opinion that Oxypolygelatin is superior to any other plasma extender now known.

  Also, it is, so far as I am aware, the only promising plasma extender to which the Government possesses a royalty-free irrevocable license. If Oxypolygelatin were to be stock-piled, rather than another material, the Government would save a significant amount in royalties;

  In addition, I think that Oxypolygelatin is considerably cheaper to manufacture than other plasma extenders that are under consideration.

  Let me mention what the royalty situation is. We, the inventors, turned our rights in the patent over to the California Institute Research Foundation, with the arrangement that no royalties were to be paid to the inventors, and that the Foundation was not to make a profit from the material, but was to be reimbursed only for expenses of administration.

  We have tried to interest a number of commercial firms in the manufacture of Oxypolygelatin. At the present time, so far as we know, only Don Baxter, Inc. of Glendale, California, is making the material.

  I may point out that Oxypolygelatin could be stock-piled very rapidly. At the present time Don Baxter, Inc. estimates that they could manufacture 100,000 units per month. A larger firm could, I am sure, very quickly reach a larger rate of production. The cost of manufacture of Oxypolygelatin may be well be less than that of gelatin itself for transfusion, because the last step, autoclaving with hydrogen peroxide present, destroys pyrogens, and there is essentially 100 per cent yield, whereas pyrogens sometimes cause batches of gelatin for transfusion to be discarded.

  Although extensive clinical testing with humans has not been carried out, it is my understanding that the material has stood up very well in these tests. Moreover, I think that clinical testing of gelatin can be considered to apply to Oxypolygelatin with considerable probability.

  I am going to be in Philadelphia on November 8 and 9, attending meetings of the American Philosophical Society. I am planning to attend a meeting of the Council on Thursday morning, and am expected, I believe, to preside at one of the sessions. If it were possible for you to allow me to make an appointment with you, perhaps I could answer any questions that you have to ask about Oxypolygelatin. If the morning of Saturday, November 10, were more convenient for you, I could meet with you then with the assurance of having no conflicting meeting of the American Philosophical Society.

  Dear Det:

  I am writing to say that I have decided not to come to the meeting of the Academy next month.

  I have not been feeling well recently - I am recovering rather slowly from an attack of influenza last month. Now my wife and I plan to go around the world, during the period November 26 to February 11. It is evident that I must get back into good physical condition, and one positive step to take, which I have taken, is to decide not to go east early in November, but rather to stay at home and rest.

  We are going around the world because of an invitation from the Government of India to spend six weeks visiting universities and research institutes in that country.

  I want to offer to serve in any capacity that I can, in connection with this trip. Does the Academy or the National Research Council, or any other organization with which you are connected, have some job to be done that I could do? I am, you know, pretty familiar with nearly all the branches of chemistry, and I have also a good bit of familiarity with physics and biology. Perhaps you would like to have a survey or report on the work in some scientific field that is being done in one of these countries, or perhaps there is a more special job to be done. Please feel free to call on me.

  My itinerary is the following: December 11-15, in Madrid; December 16-20, in Athens; December 21-22, in Cairo; December 23-January 31, in India, probably visiting many places; February 1-5, in Bangkok; February 6 in Hongkong; February 7-10, in Tokyo.

  I may say that I would be especially interested in having a job to do in Japan. There is a great deal of activity in the chemical field in Japan; in particular a great deal of work on the determination of the structure of crystals with x-rays is being done there, and, indeed, there is an active group working on the structure of proteins. It would be, I think, quite valuable for me to have first-hand contact with workers in this field, and to find out just what is going on. I think that the work that Professor Corey and I, and our younger colleagues, are doing here

  Dr. Bronk

  on the problem of the structure of proteins is pretty important, and that the possibilities for the future are great. The work might be helped significantly by information about Japanese work that I could obtain through personal contact.

  I understand that it is still necessary to have permission of the Military Occupation authorities to visit Japan, and I am hoping to apply for this permission. If I had official backing - of the Academy or Research Council, for example - the chances of my obtaining permission would be greatly increased.

  Thank you for your letter of October 16, in which you mention the celebration of American Soviet Friendship Week in the San Francisco Bay Area, early in November.

  I hope very much that a permanent time of international good feeling and of peace can be instituted, and that the world will soon have passed through the present troubled period.

  I have felt deep concern about the state of the world, and especially in respect to science, during recent months. It seems to me that the field of science should be truly international, and that there should be no restrictions on the dissemination of knowledge and on the movement of scientists. Also, it is clear that science must be allowed to develop without artificial restrictions or governmental interference - there is no other way in which the truth can be discovered.

  The World Chemical Conclave was held in New York last month. I know that Russia was invited to send representatives - but none came. I attended the previous International Congress of Pure and Applied Chemistry, in London, in 1947. Three Russians appeared there, at the last moment, and without warning - there had been no statement about their attendance sent in ahead of time, to permit the titles of their papers to appear in the program. These Russian chemists presented their papers, but they did not talk with the other chemists attending the meeting, so far as I am aware. I wish that more scientific intercourse could take place.

  Also, I have been deeply concerned about the attack that has been made upon my own work. I told you about the published report to the Academy of Sciences, by a committee of eight chemists, in which my work and the theory of resonance are severely criticized, and it is essentially forbidden to Russian chemists to make use of the theory. I have since learned that this attack has been going on since 1949 - it began with a virulent article in the Journal of Philosophy. Several other papers along the same line have since appeared.

  I have not been able to gain any understanding whatever of the meaning or cause for such an attack. I would be grateful to you for your ideas, and any information you can give me.

  The literature on the above subject is very meager but interestingly enough what little is available is in good empirical agreement with the relative rates of hydrolysis of the peptide bonds of those peptides which have been isolated from lysosome: in the case of a regular structure such as that postulated for collagen and gelatin, one might anticipate the breaking of particular bonds.

  I should like to present the information in the literature and in our own data for your consideration because I believe that if it were possible to decide what are the causes of the differences in the ease of hydrolysis of various peptide bonds it would greatly aid our studies of protein structure.

  Literature

  In studying the hydrolysis of gramicidin, Christensen (J. Biol. Chem., 151, 319 (1943) and 154, 427 (1944)) observed that the hydrolysis of valylvaline was very difficult even in refuxing hydrochloric acid. Synge (Biochem. J., 44, 542 (1949)) substantiated this work but earlier (Biochem. J., 39, 351 (1945)) he had studied the relative rates of hydrolysis in acid at 37° of a number of peptides. He found that glycyl peptides (for example, glycylleucine, glycylglycine was more difficult and valylglycine was very difficult to hydrolyze. Form these results, the R group attached to that amino acid which contributes the –CO- to the peptide bond appears to play an important role. Christensen and Hegsted (J. Biol. Chem., 158, 593 (1945)) made a further study of the hydrolysis of the various peptide bonds in gramicidin at 37° and at refluxing temperatures and concluded that the release of valine is slow, that the release of tryptophan is more rapid than that of the others except at 37°, that linkages with alanine are relatively stable at both temperatures, and that, when leucine contributes –CO- to the peptide bond, the bond is relatively stable to boiling.

  Desmielle and Casal (Biochim. Biophys. Acta, 2, 64 (1948)) have demonstrated that a very labile bond results when serine or threonine contributes –NH- to the peptide bond and they have evidence that the following shift takes place under the influence of strong acid: [Figure of Chemical Reaction Step]

  Our Work

  In our attempt to produce longer end peptides from DNP-lysozyme a study was made of the nature of the peptide mixture which is obtained by refluxing in 6 N hydrochloric acid for various periods. The results are shown in Figure 1. Analysis of the peptides established the sequence pictured in Figure 2. The conclusion may be drawn from these data that the relative rate of fission of the bonds is

  Partial hydrolysis of each of the peptides substantiates these conclusions.

  It has also been possible to recognize seryl and threonyl peptides: in one- experiment, lysozyme itself was partially hydrolyzed and the partial hydrolysate was dinitrophenylated. From the mixture, seryl and threonyl peptides were isolated end their composition may have been seryl (leucine, alanine, glycine) in which the sequence of leucine, alanine, and glycine is undetermined and threonylaspartic acid.

  Remarks

  The information in the literature would lead us to expect that Bond 2 would be a relatively stable one and Bond 4 a relatively labile one and this is found to be the case. No data about bonds like 1 and 3 seem to be available. Bond 5 may be very stable or Bond 4 may be a very labile one if the fifth amino acid should be serine or threonine or perhaps tryptophan.

  The isolation of seryl and threonyl peptides provides further proof of the work of Desnuelle and Casal. If the seryl peptide should be seryl (leucine, alenine) glycine, this fact would indicate again the lability of bonds in which glycine provides –CO-.

  In attempting to explain the observed facts, one immediately thinks of the possibilities of steric hindrance. The use of the models shows that in an extended polypeptide chain the R group of an amino acid in certain positions could protect one side of the bond to which its amino acid contributes -CO- but that the other side would be open to the hydrolyzing medium unless protected by the R group of the amino acid which provides -NH- to the bond. On this basis, the stability of valylglycine toward hydrolysis is difficult to explain. Of course, if hydrolysis requires the approach of the hydrolyzing molecule from a certain direction, the protection furnished by one R group may have significance.

  On the other hand, in the 3,7—residue helix the R group can provide no protection to that bond to which the amino acid contributes -CO- but it can protect, that to which it contributes -NH-.

  Perhaps the differences in the ease of hydrolysis of the various peptide bonds is associated with the resonating character of the bond but it is somewhat difficult to see how the R groups would influence it.

  I would greatly appreciate any comments which you would care to make on this problem.

  Titanium compounds isomorphous with high-speed steel carbide.

  A few days ago I mentioned to Mr. Pietrokowsky my interest in the work of Karlsson on the iron-titanium-oxygen compounds, and similar compounds, with the Fe3W3C structure. He said that you had attempted to repeat this work, without success.

  I would be glad to know what results you obtained in this investigation. Dr. Ewing is making a study of the electronic structure of these substances.
